In published research, FOXO4-DRI (Proxofim) has been investigated for mechanisms including: FOXO4-DRI is a cell-penetrating peptide antagonist that targets the FOXO4-p53 interaction in senescent cells. In senescent cells, FOXO4 abnormally accumulates in the nucleus, binds to p53. FOXO4-DRI displaces native FOXO4 from p53, freeing p53 to relocalize to mitochondria, initiate apoptosis specifically in senescent cells, while sparing healthy cells.
